The BlotCycler Touch is a versatile and powerful blotting processing system that can be used for a variety of applications, including antibody titration. The BlotCycler Touch has the capability to run six different primary and secondary antibodies simultaneously. Due to this unique feature, six different concentrations of primary antibodies can be tested to determine the optimal concentration. Due to the cost of primary antibodies and there frequency of use in Western Blotting a definite cost savings can be realized.

We recommend following these steps:

  1. Prepare your samples. Dilute your primary antibody to a range of concentrations. It is good practice to start with a high concentration and dilute down by a factor of 2 or 3 each time.
  2.  Load your samples onto the BlotCycler Touch and run the appropriate blotting protocol.
  3. We recommend running all steps at 4 degrees Celsius
  4. Detect your proteins. Once the blotting protocol is complete, use your preferred method to detect your proteins.
  5. Analyze your results. Once your proteins have been detected, you can analyze the results to determine the optimal antibody concentration. Look for the band that is darkest and most specific for your protein of interest.

Here are some additional tips for using the BlotCycler Touch for antibody titration:

  • Use the proper blocking buffer for the experiment. Blocking buffer helps to prevent non-specific binding of the antibody to the blot.
  • Make sure to use a positive control. A positive control is a sample that is known to express the protein of interest. This will help you to ensure that your blotting protocol is working properly.
